IMO, if you get as far as Castroe, you don't need to be told what to do with the e. The question mark in the clue is meant to indicate something is not quite exact.

No, it doesn't. That's not what question marks are for in crosswords. It means there may be a play on words, but 'ro' isn't a word. Ro would have to be defined by something like 'endlessly'.
